Custom Boss Help

I'm still learning the capabilities of this engine, and I have a couple questions:
 

Is there a way to make an boss NPC that is only attachable in a certain direction (i.e. the back), like a weak spot? 
 

Is it possible to give an NPC a rout? 
 

I saw a forum post that said a guy programmed his bosses. How can I do that?
